When you say "tile", are we talking about conventional WMS or WMS-C?
If you mean conventional WMS, where a bounding box request is sent to a server that composes an image for the area, these kind of requests cannot be cached with Squid, etc.
I don't have much experience with WMS-C, but with my understanding of how it works, it should work with conventional HTTP caching software like Squid, etc.
You may want to look at Tilecache[1]. It can cache conventional WMS requests and serve up that cache as WMS-C server/TMS, provided the app you're using supports those.
