Yes, this can be done in OSM, or rather OpenLayers.

Have a look at the javascript code and especially the KML files that 
come with http://garmin.na1400.info/routable.php for the transparent 
areas with different colours. Then have a look at 
http://yournavigation.org for an application that uses markers in 
OpenLayers.
