First you find out the URL of the tile: In Firefox I just right-click on the tile I'm interested in and select View Image. 

That gives you a URL like this
	http://a.tile.openstreetmap.org/14/13699/7522.png

To mark it as dirty, just add /dirty on the end of the URL in the address bar, and hit return:

	http://a.tile.openstreetmap.org/14/13699/7522.png/dirty

You get the message "Tile submitted for rendering", and it will go away and re-render the tile. 

It might take a few minutes, but usually this works pretty quickly
